Incomplete Microtubules Observed in Mammalian Blood Platelets during Microtubule Polymerization

and 0°C. In both cases the process was reversible (1), 1 the microtubules reappearing when the cold treatment was discontinued. This work deals with the cytoplasmic microtubules in mammalian blood platelets. Unlike the microtubules of the axial filament complex of cilia and flagella, these microtubules are properly preserved only by glutaraldehyde fixation. In platelets the microtubules are arranged in a marginal bundle (3 5) which is thought to be responsible, at least in part, for the maintenance of the lenticular shape of the circulating blood platelet. The report presents some observations on cytoplasmic microtubules of the marginal bundle of mammalian blood platelets subjected to changes in environmental temperature, and shows that, like cytoplasmic microtubules in other cells, platelet microtubules disappear when platelets are placed in the cold, and that they reappear when the platelets are warmed again. Observations are presented which give indications of how microtubules are formed. Changes in the shape of platelets concomitant with changes in the marginal bundle of microtubules will be dealt with in detail in a later report.

P-121: Assessment of Microtubule and Nuclear Status at Different Intervals of Bovine In Vitro Oocyte Maturation
Background: Mammalian oocyte undergoes a series of structural nuclear modulations during maturation in order to obtain full competence to support fertilization and early embryonic development. Microtubules are major cytoskeleton components and have pivotal modulators for chromosomal movement, and it has been shown that partial or immature spindle organization may compromise correct ploidy of th...
متن کاملThe Effect of the Crocus Sativus L. Carotenoid, Crocin, on the Polymerization of Microtubules, in Vitro
Objective(s): Crocin, as the main carotenoid of saffron, has shown anti-tumor activity both in vitro and in vivo. Crocin might interact with cellular proteins and modulate their functions, but the exact target of this carotenoid and the other compounds of the saffron have not been discovered yet. Microtubular proteins, as one of the most important proteins inside the cells, have several functio...
متن کاملUnique functional characteristics of the polymerization and MAP binding regulatory domains of plant tubulin.
An understanding of the regulation of microtubule polymerization and dynamics in plant cells requires biochemical information on the structures, functions, and molecular interactions of plant tubulin and microtubule-associated proteins (MAPs) that regulate microtubule function. We have probed the regulatory domain and polymerization domain of purified maize tubulin using MAP2, an extensively ch...
متن کاملJCB: Report
Mammalian platelets are small cell fragments with neither nu cleus nor microtubule organizing center. They circulate in the blood stream to survey vascular integrity. In their quiescent state, they have a flat, discoid morphology caused by a micro tubular ring structure, called the marginal band (MB), in the pe riphery of resting platelets (White and Rao, 1998). Endothelial damage will induc...
متن کاملAltered microtubule equilibrium and impaired thrombus stability in mice lacking RanBP10.
The crucial function of blood platelets in hemostasis is to prevent blood loss by stable thrombus formation. This process is driven by orchestrated mechanisms including several signal transduction cascades and morphologic transformations. The cytoplasmic microtubule modulator RanBP10 is a Ran and β1-tubulin binding protein that is essential for platelet granule release and mice lacking RanBP10 ...
